Scientists Identify Mutation Hotspots in Human Genome

Researchers have discovered specific regions in the human genome that are more prone to developing mutations. These mutation hotspots occur at transcription start sites, where RNA polymerase ‘opens’ the DNA to read and copy instructions. Geneticist Donate Weghorn from the Centre for Genomic Regulation in Spain emphasizes the importance of these sequences, which are crucial for understanding genetic diseases.

While most mutations are harmless, some can be detrimental and even inherited by offspring. Approximately 300 million people worldwide are affected by rare genetic disorders, highlighting the significance of studying mutation susceptibility in the human genome. Damage to the genome increases during transcription, where DNA is copied into RNA molecules.

Through extensive analysis of human genome datasets, researchers found a strong mutation hotspot near transcription start sites in individuals with inheritable mutations. This hotspot was absent in de novo mutation studies but reappeared in mosaic mutation data, which occur during early embryonic development.

According to Weghorn, there is a blind spot in current mutation studies that overlooks the impact of mosaic mutations. By examining all three datasets together, researchers uncovered the mechanisms behind the vulnerability of transcription start sites to mutations.

The findings shed light on the origins of DNA mutations and may improve studies on genetic conditions that rely on de novo mutation data. The research has been published in Nature Communications.
