The enigmatic ‘vampire squid from hell’ has recently unveiled its largest cephalopod genome ever sequenced, a whopping 11 billion base pairs – more than double the size of the largest squid genomes. Despite not being a true squid, Vampyroteuthis infernalis has maintained a squid-like chromosomal architecture, shedding light on its deep evolutionary history shared with modern octopuses and squids.

This peculiar creature, considered a living fossil dating back 183 million years, is the last surviving member of an ancient lineage that has since disappeared. Its genetic makeup offers insights into the mysterious origins of cephalopods, revealing vestiges of shared traits with squids, octopuses, and cuttlefishes.

Genomicist Oleg Simakov from the University of Vienna notes, “The vampire squid sits right at the interface between octopuses and squids. Its genome unveils deep evolutionary secrets on how two distinct lineages could emerge from a common ancestor.”

Despite its elusive nature and deep-sea habitat exceeding 600 meters, researchers were able to sequence the vampire squid’s DNA from a specimen accidentally caught by the Tokai University research vessel. Surprisingly, the genome size ranges from 11 to 14 gigabases, dwarfing other cephalopod genomes like the longfin inshore squid and the common cuttlefish.

A significant portion of the genome comprises repetitive elements, contributing to its immense size without adding new coding sequences. Comparative analysis with other cephalopod genomes highlighted the unique genetic makeup of the vampire squid, positioning it as a key player in deciphering cephalopod evolution.

The findings suggest that octopuses underwent rapid chromosomal mixing early in their evolutionary history, while vampire squids retained a more stable chromosomal structure despite genome expansion. This makes the vampire squid a potential Rosetta Stone for understanding the earliest stages of cephalopod evolution.

Genomicist Emese Tóth of the University of Vienna remarks, “The vampire squid retains a genetic heritage that predates both squid and octopus lineages, offering a direct glimpse into the earliest cephalopod evolution stages.”

This groundbreaking research, published in iScience, showcases the vampire squid’s pivotal role in unraveling the complex evolutionary history of cephalopods.
